My most memorable moments fitting the criteria were: PvCvW and Magda challenge beasting the Resurrection Chamber and making it to the jury despite being first voted out.

And in Pokemon: Colosseum with Magnemite getting increasingly frustrated at being called "a magnet with bits attached".

Oh wow, we've reached the point where nostalgia is nostalgic

The things that are memorable now are different than what was memorable six years ago. I'm surprised by how many experiences with challenges live in my head rent-free, as opposed to big moments socially. Driving the ship forward, the absurd grid challenges in Mario Bros Survivor, the fairy logic puzzle in Hyrule, the very first experiences with JigsawPlanet, 3-Player Chess in Pokemon Colosseum, etc. Those are the clear ones for some reason.

In terms of challenges:

Jigsaw Planet definitely lives in my head to this day.
The Egg-Throwing challenge in Eon.
Broth of the Weld and Shield Surfing from PvCvW
Secret Club Secrets from PvCvW

Though most of those the iconic aspect came from the banter/conversations that came up during the challenge? Egg-throwing cross-tribe banter, bonding over music sharing and coaching each other through shield surfing challenge, and then fifteen players scrambling to figure out what tribe they're in during Secret Club Secrets.

But also the tension of Broth of the Weld in terms of the timing in the game (Resurrection Chamber re-entry) made it so much more tense.
